Artifact ...

Last week I had a cool result - but this week after looking at all the controls it would seem like that amazing result is an artifact.

Oh well ...

Instead of showing you that great result, I'll post the remains of another cell that exploded after microinjection. Note that quite a bit of the membrane stayed behind giving the cellular debris a funny appearance.

When I was a tech, I tried microinjecting dye into NRK cells. I was never very good at it--I always gouged the hell out of the cells or smashed the tip of the micropipet into the plate. Clearly I had problems with depth perception on the microscope. Anyway, I gave it up after awhile and we decided not to do those experiments (I guess nobody else in the lab wanted to try their hands at dye injection).

Damn artefacts! Although, I recently did a set of expts that went like this: 1) Wow!!! Great staining of interesting structures!!! 2) Shit. Completley non-specific antibody that stains untransfected cells and about 89 bands on a Western blot. 3) Hang on ... Random non-specific Ab stain colocalizes with specific marker for what I looking for anyway!!!
So, you never know.
